Description:
Definition: An enterocyte of the human colon (large intestine) expressing carcinoembryonic antigen-related cell adhesion molecules 1, 3, 5, and 6 (CEACAM1, CEACAM3, CEACAM5, CEACAM6) , also known as CD66acde.
Localization: Predominantly found in the colon at the top of the mucosa, representing mature enterocyte populations.
Characteristics: These cells are characterized by the expression of CD66acde, indicating a mature state and specific functional properties within the colonic epithelium.
Significance: CD66acde is involved in cell adhesion and intracellular signaling. Its expression on enterocytes signifies maturation and possibly roles in barrier function and cell-cell communication (CD66 is also present on neutrophils but CD66 + Pan cytokeratin will indicate epithelial while CD66 + CD45 will indicate neutrophils).
Note on CEACAM Subunits:
In our study, CD66acde (CEACAM1, CEACAM3, CEACAM5, CEACAM6) were the specific subunits used to identify these enterocytes.

Markers Associated with Immune Cells:
While CD66a is traditionally associated with immune cells (e.g., neutrophils), its expression on enterocytes marks unique epithelial subsets with specific functions or maturation states.
Disease Association:
Hickey et al 2023. Nature 619 (7970): 572–84. study focused on healthy intestinal tissues. The expression of this marker in enterocytes within healthy samples suggests that they are part of normal physiological processes rather than disease states.

We identified this enterocyte type to capture the heterogeneity within the intestinal epithelium:
CD66a-positive enterocytes denote mature enterocytes prevalent in the colon.
